What is Aminophylline and how does it work?

Pharmacological effects of aminophylline

Aminophylline is a double salt of theophylline and diethylamine. The main pharmacological effects of theophylline come from theophylline. Ethylenediamine enhances its water solubility.
① Relax bronchial smooth muscle, and also relax various smooth muscles such as intestine and biliary tract, and also relieve congestion and edema of bronchial mucosa.
②Increase cardiac output, expand output and input renal arterioles, increase glomerular filtration rate and renal blood flow, and inhibit the reabsorption of sodium and chloride ions in the distal renal tubules.
③Increase the contractility of isolated skeletal muscle; in the case of chronic obstructive pulmonary disease, improve the muscle contractility. Theophylline increases ventilatory insufficiency during hypoxia and is thought to be because it increases the contraction of the diaphragm, and its role in this aspect exceeds that of the respiratory center.

How Aminophylline uses for?

Aminophylline indications

1. Bronchial asthma and asthma-like bronchitis, combined with β receptor agonists can improve the efficacy. In persistent asthma, the combination of aminophylline and adrenal cortex hormones is often used for treatment.
2. Treatment of acute cardiac insufficiency and heart failure asthma (cardiogenic asthma).
3. Biliary colic.
4. Treatment of sick sinus syndrome and sinus block.

Usage and dosage

1. The usual dosage for adults is 0.1~0.2g per day, 0.3~0.6g per day; the maximum amount: 0.5g per time, 1g per day.
For intramuscular injection, 0.25~0.5g once, 2% procaine hydrochloride should be added.
Intravenous injection, 0.25~0.5g once, 0.5~1g per day, 25~100 mg diluted with 5% glucose injection to 20~40ml, the injection time should not be shorter than 10 minutes.
Intravenous infusion, 0.25~0.5g once, 0.5~1g a day, diluted with 5~10% glucose solution and then slowly instilled.
For injection, the maximum amount is 0.5g once and 1g per day. Rectal administration, usually before going to bed or after going to bed, 0.25-0.5g once, 1-2 times a day.

2. The usual dose for children is taken orally, according to the body weight of 4-6 mg/kg, 2 to 3 times a day. Intravenous injection, one time according to body weight 2~4 mg/kg, diluted with 5-25% glucose injection, and slowly injected.

What are the Aminophylline side effects?

(1) Common adverse reactions are: nausea, stomach upset, vomiting, loss of appetite, headache, irritability, and irritability can also be seen.
(2) When aminophylline is poisoned, it is manifested as arrhythmia, increased heart rate, muscle tremor or epilepsy. Due to irritation of the gastrointestinal tract, bloody vomit or tarry stools can be seen.

Aminophylline drug interactions

1. Dilute hydrochloric acid can reduce its absorption in the small intestine.
2. Acidic drugs can increase its excretion, while alkaline drugs reduce its excretion.
3. Cimetidine, erythromycin, clindamycin, lincomycin, and tetracycline can reduce the clearance rate of aminophylline in the liver and prolong t1/2, so the blood concentration can be higher than normal. Prone to poisoning.
4. Sodium phenytoin accelerates its metabolism and reduces the blood concentration, so the dosage should be increased accordingly.
5. When combined with propranolol, the bronchodilator effect of aminophylline may be inhibited.
6. Combined use with lithium can accelerate the excretion of lithium by the kidneys, so the curative effect of lithium decreases.
7. During intravenous drip, avoid the compatibility with vitamin C, corticotropin, norepinephrine, and tetracycline hydrochloride.

Aminophylline use as a veterinary medicine

Aminophylline has the strongest relaxing effect on bronchial smooth muscles, which can dilate the bronchus, increase lung capacity, and have a longer-lasting effect, especially for bronchial spasticity. In addition, aminophylline also expands coronary arteries. Increase the blood supply of the myocardium and strengthen the contractility of the heart. It is mostly used for emphysema of horses and cattle, as well as relieving asthma of dogs with pulmonary congestion (cardiac wheezing) caused by heart failure.

Limitations of BALLYA Aminophylline Test?

BALLYA Aminophylline Test is a qualitative test kit. It’s only for screening purpose. If have positive cases or suspected case, maybe use other detection method to make a further detection, such as, ELISA, PCR, qPCR, etc.

Conclusion

As a pharmaceutical feed additive, aminophylline can significantly promote weight gain and increase feed conversion rate, and is widely used in the field of livestock and poultry breeding. However, many people do not pay attention to the dosage and usage of aminophylline, which leads to the residue of aminophylline in food. Aminophylline is harmful to the human body and should be avoided as much as possible.
The BALLYA Aminophylline Test provided by BALLYA can effectively detect the aminophylline in the samples. Simple operation, short detection time and high accuracy are the characteristics of this product.
