Introduction
The Brucella Ab Rapid Test Kit uses a competitive method to qualitatively detect brucellosis antibodies in the serum and whole blood of animal species including bovine, porcine and ovine.

Test procedure
1. Pipetting 0.5-1 ml of blood, centrifuge for 3-5 minutes at 3000 r/m to separate the serum. It can also be left standing overnight at 4ºc to allow the serum to separate out by itself. Or test the whole blood without preprocessing.
2. Let the test card at room temperature, take out and place it on the flat desk.
3. Take 5ul (as the black mark on the capillary) of the serum or whole blood into the sample well, and immediately add a drop of diluent by the diluents dropper.
4. Please wait for 5 minutes.
5. Add two more drops of diluents.
6. You will see a burgundy liquid flowing through the screening hole. Read the result for 10-15 minutes. The result after 15 minutes is invalid.

Brucella Ab Rapid Test Kit detection principle

The kit uses double antibody one-step sandwich enzyme-linked immunosorbent assay (ELISA). To the coated microwells pre-coated with antibody, add the specimen, standard and HRP-labeled detection antibody in sequence, incubate and wash thoroughly. The color is developed with the substrate TMB, which is converted into blue under the catalysis of peroxidase, and into the final yellow under the action of acid. The color depth is positively correlated with the sample. The absorbance (OD value) was measured with a microplate reader at a wavelength of 450 nm, and the sample activity was calculated.

Sample collection of Brucella Ab Rapid Test Kit

1. The sample must not contain sodium azide (NaN3) because sodium azide (NaN3) is an inhibitor of horseradish peroxidase (HRP).
2. Extract the specimens as soon as possible after collection.
3. Plant extracts or other related samples: centrifuge at 1000 x g for 20 minutes, and take the supernatant for testing.
4. Preservation: If the sample is not tested in time after collection, please aliquot it in one dose and freeze it at -20 ℃ to avoid repeated freezing and thawing. Thaw at room temperature and ensure that the sample is thawed evenly and fully.

Operation steps of Brucella Ab Rapid Test Kit

1. Take out the required slats from the aluminum foil bag after equilibrating at room temperature for 20 min. The remaining slats are sealed with a ziplock bag and put back at 4 °C.
2. Set up standard wells and sample wells, add 50μL of standard products with different concentrations to the standard wells;
3. Add 10 μL of the sample to be tested to the sample well, and then add 40 μL of the sample diluent; blank wells are not.
4. In addition to the blank wells, add 100 μL of horseradish peroxidase (HRP) -labeled detection antibody to each of the standard wells and sample wells, seal the reaction wells with a sealing plate, and incubate in a 37 °C water bath or incubator 60min.
5. Discard the liquid, pat dry on the absorbent paper, fill each well with washing liquid, let stand for 1min, shake off the washing liquid, pat dry on the absorbent paper, repeat the plate washing 5 times in this way (you can also use the plate washing machine to wash the plate).
6. Add 50 μL of substrate A and B to each well, and incubate at 37 °C in the dark for 15 minutes.
7. Add 50μL of stop solution to each well, and measure the OD of each well at 450 nm within 15min.
